PhiNPQ, NPQ(T) and FvP/FmP

By Weizhen Liu

MultispeQ v1.0 gives us both PhiNPQ and NPQ(T) values. Based on my reading of Tiet et al. 2017, PhiNPQ needs to be estimated after dark-adaptation but NPQ(T) does not. Using MultispeQ in the field with less than 1 minute measurement of each plant leaf, I think the leaf does not go through dark-adpation process before taking measurement. Can I say that NPQ(T) is more accurat... read more

Missing a lot of spreadsheet columns?

By David Alm

Greetings, I haven't used instruments since last Spring. I just started using them again, and only basic data are in the spreadsheet (Phi's, chl,..) but these are missing: Proton Motive Force: ECSt, vH+, gH+ Absorbance at: 450, 535, 605, 650, 730, 850, 880, and 940nm. What am I doing wrong? I looked all over the GUI for a widget to include more columns, but couldn't find.... read more

How can we use internal light LED as PAR or ambient light

By Habib-ur-Rehman Athar

From my project photosynthetic variations in canola under salt stress, I can see greater Phi2 values of salt stressed plants than their respective Control plants. Probably, it may be due to difference in PAR. (PAR ranges from 900-1200 uE) Since values were taken in wire-net house, it is very difficult to maintain PAR. Is it possible to normalize Phi2 values as function of P... read more

Actinic light -MultispeQ V2.

By Laura Briers

Hi, I wonder whether you could help. We have just ordered a MultispeQ V2. I wanted to know what the actinic light of the MultispeQ is? or is the actinic light used determined by the ambient PAR? I had read that the PAR light received by the PAR sensor is recreated within the chamber via amber LED's. If the actinic light is determined in a macro, is there anyone to edit thi... read more

macro updating project output?

By Jonathan R Pleban

Hi Ive recently updated a macro: v1 NPQ 5 min light acclimation 20 dark recovery JRP This macro is used for protocol: v1 - NPQ 5 minutes light 20 min dark recovery - 800 The protocol is currently used in the NPQ recovery B. rapa drought project. In the past when i've changed macro's the new outputs are available right away; now I am getting no output in project data at a... read more

Using a macro to calculate stomatal conductance.

By Patrick S.

I'm looking to use the MultispeQ to measure stomatal conductance since existing protocols claim that they can estimate stomatal conductance via changes in air humidity and leaf temperature. I was looking at using the following protocol: https://photosynq.org/protocols/rides1_3_1 The corresponding macro is: https://photosynq.org/macros/rides1_3_1 I was wondering how I can... read more

During measurement readings using Photosynq device there is a Checksum error that continuous appears on my Samsung TabA model SM-T280. Once this appears, I have to re-take the measurement. Other times, I have closes the program and re-open it. The error message persists in both instances. Does anyone have thoughts on what might be the issue? Thanks.

PhiNO

By Craig Whippo

My students are looking at how photosynthesis is changing over the course of fall phenology. Junker and Ensminger show that phiNO increases as chlorophyll is lost in maple leaves. This sounds reasonable to me based on the relationship between NPQ and pmf. However, my students are getting data from the multispeQ showing that phiNO is lower in yellow leaves. I don't under... read more

What do raw traces actually mean

By Alejandro Morales

Hi, This may be a silly question but I cannot find any explanation on the website or the wiki from the Github repo. What exactly do the numbers in the raw traces mean? From some simple tests I have figured out they depend on both brightness and length of the pulses but I don't know what physical quantity is behind these numbers. My end goal is to calculate the fraction of... read more

Why did I get negative values for leaf thickness?

By Weizhen Liu

I used Leaf Photosynthesis MultispeQ V1.0 protocol to measure different genotypes of corn leaf blades. When exploring data after measurements, I found that majority thickness values were negative. Please see the distribution figure attached. The protocol " Leaf Photosynthesis MultispeQ V1.0" described that it measures Leaf Thickness in mm. I do not know why I obtain negativ... read more

Hi, I was wondering how to change the settings on the desktop app so that I can collect data without access to the internet. I am using a USB connection from the laptop to the instrument. I saw in the tutorials there is a way to do so for the mobile app, but can't seem to find it in the desktop app. Thanks, Naomi
